Abstract

Concerns about antibiotic residues in milk and emergence of bacterial resistance necessitate exploration of alternative therapeutic strategies to antibiotics for treatment of mastitis. The study was carried out to investigate the antibacterial activity of ethanol extracts Ya-Sa-Marn Phlae (THR-SK010), its herbal components including Curcuma longa Linn., Areca catechu Linn., Oryza sativa Linn., and Garcinia mangostana Linn., as well as their representative chemical constituents including catechin, alpha-mangostin, and curcumin against Staphylococcus spp. isolated from mastitis cows. Additionally, testing the susceptibility of the pathogen to antibiotics as well as mechanisms of action of the agents on the representative clinical isolates were evaluated. Most of tested isolates obtained from mastitis were resistant to penicillin and ceftazidime. The extracts of Ya-Sa-Marn-Phlae, Garcinia mangostana Linn., and alpha-mangostin exhibited remarkable antibacterial effects against all tested staphylococcal isolates. At 4MIC, the agents additionally caused lysis, as determined by measurement of the optical density at 620 nm. Loss of 260-nm-absorbing materials also occurred after treatment with the extracts of Ya-Sa-Marn-Phlae, Garcinia mangostana Linn., and alpha-mangostin at 4MIC. The anti-staphylococcal effects were confirmed with both scanning and transmission electron microscopes. Moreover, Ya-Sa-Marn-Phlae, Garcinia mangostana Linn., and Curcuma longa Linn. extracts inhibit biofilm formation of coagulase positive staphylococci (BCPS) 31 as well as Staphylococcus epidermidis ATCC 35984. The current study indicated that Ya-Sa-Marn-Phlae had strong antibacterial activities and anti biofilm abilities against staphylococci isolated from bovine mastitis similar to that of Garcinia mangostana Linn., alpha-mangostin, and Curcuma longa Linn.
