Abstract

Nowadays, antimicrobial resistance has been widely spread because of using antibiotics to control pathogenic bacteria causing fish diseases. However, the residues of antibiotics possess several side effects on fish bodies, aquatic ecosystems, also consumers. Probiotic is an alternative way for preventing and protecting the aquatic system, including the health of fish, leading to the consumer. Nevertheless, probiotics need prebiotics as food to feed them for their efficient processes. This study aims to extract and examine the prebiotic properties of Saba banana extract in the growth promotion of probiotics bacteria in a group of lactic acid bacteria (LAB), including Weissella confusa, Lactobacillus lactis, L. plantarum ATCC14917 and L. casei ATCC7469. In the present study, the powder was prepared by soaking in 50% ethanol. The mixture was placed in a shaking incubator. After that, the mixture was filtered with a double layer. Then, centrifugation was done for 30 minutes. The solutions was eliminated ethanol with rotary vacuum evaporation and powdered by freeze dryer. The minimum inhibitory concentration (MIC) of banana flesh extract (BFE) and banana peel extract (BPE) against Vibrio spp. was determined by the broth microdilution method. The concentration ranged from 1.024 to 32.768 mg/ml. After incubation, the MIC values were evaluated by adding 0.1% resazurin solution. Moreover, the result of chemical analysis such as reducing sugar, total carbohydrate, phenolic content, and flavonoid content showed as 144�0.25 mg/g, 47.47�1.65 mg/g, 310.44�16.86 mg GAE/g and 787.94�61.42 mg QE/g, respectively, for BFE and 67.91�1.59 mg/g, 94.87�2.26 mg/g, 744�15.61 mg GAE/g and 1,220�64.26 mg QE/g, respectively, for BPE. In addition, % acid hydrolysis and % enzymatic hydrolysis were analyzed and demonstrated the complacent results about prebiotics as well. All the results from prebiotic properties testing of BFE and BPE exhibit a great potential to be used as sustainable prebiotics to promote food safety and environmentally friendly aquaculture.
